Isner's most famous victories were against Roger Federer in the Davis Cup encounter in February 2012, top seed Novak Djokovic at the 2012 Masters 1000 tournament in Indian Wells, Andy Roddick in the 2009 US Open, and against Nicolas Mahut at the 2010 Wimbledon Championships where he played the longest ever professional tennis match, which lasted 11 hours and 5 minutes over the course of three days. He also played in the 10th-longest match ever—a 5 hour 41 minute match in the second round of the 2012 French Open.

Isner had the best season of his career so far in 2012. He defeated a World No. 1 tennis player for the first time and played a key role in the Davis Cup for the US.

John Isner, known for his ability to hit aces, was the ATP World Tour leader in aces in 2012.

John Isner believes Rafael Nadal has the greatest muscular endurance on the ATP Tour. He finds Roger Federer to have the best balance and Novak Djokovic to be the most flexible. Isner believes Nadal is the quickest and Federer to have the best footwork.
