Serena Williams entered China Open 2009 in October seeded second. She defeated Kaia Kanepi in the first round. In the second round, she beat Ekaterina Makarova 6-3, 6-2 and became World No. 1 replacing Dinara Safina. In the third round, Serena lost to Nadia Petrova 4-6, 6-3, 6-7 (5-7). Serena Williams remained World No. 2 for two weeks and was dropped to No. 2 again in the rankings released on October 26, 2009.

In the season ending Sony Ericsson Championships 2009, Serena Williams was seeded second and is placed in the Maroon group alongside Svetlana Kuznetsova, Elena Dementieva and Venus Williams. She defeated Svetlana Kuznetsova 7-6 (8-6), 7-5 in her first group match. In the second match, Serena played sister Venus Williams. Serena dropped the first set, but won the match 5-7, 6-4, 7-6(4). On the same day, Dinara Safina retired from the tournament due to a back injury which means that Serena Williams will end the season as World No. 1. Serena defeated Elena Dementieva by 6-2, 6-4 in her final round robin match and became the first player to qualify for the semifinals in the tournament.

In the semifinal, Serena Williams defeated Caroline Wozniacki 6-4, 0-1 as Wozniacki retired in the second set with an abdominal strain. In the doubles event, She and sister Venus Williams lost to Nuria Llagostera Vives / Maria Jose Martinez Sanchez 2-6, 6-4, 10-8 on their debut in Women's doubles in the tournament.

Serena Williams has withdrawn from the U.S. team for the Fed Cup final against Italy, deciding she needs rest after a long season.
